- Cells grown in culture medium were injected in a chip (as in Additional file 4C) to determine the appropriate focal distance for fluorescence quantification. The observation area was then moved to the elastomer to measure its autofluorescence. Culture medium between two glass coverslips was used as a control (glass). Averages of the fluorescence over more than 2000 pixels along a diagonal line scan with standard deviations are shown for the indicated excitation wavelengths. Note that potential autofluorescence from the elastomer would only minimally affect imaging, as by design, there is no elastomer in the light path (see Fig. 2a)
