  1. Quantitative RT-PCR analysis of NFATc1 mRNA expression in RAW264.7 cells. Cultures were treated with RANKL, RANKL and IL-10 or RANKL, IL-10 and PMA for 24 hours and total RNA extracted. Quantitative PCR was performed on a Techne Quantica PCR machine using the DNA binding dye SYBR-green and specific primers for murine NFATc1 and β-actin. For each sample the NFATc1 copy number was expressed as the relative copy number normalised to 106 β-actin copies. Samples were analysed in triplicate and are expressed as the mean of three separate experiments. * p < 0.05 versus RANKL group.
